1. Preparation of BrdU stock solution

3. Discard the culture medium and wash the slides 3 times with PBS.

 

4. Methanol / acetic acid fixation for 10 min.

 

5. The endogenous oxidase was inactivated by 0.3% H2O2-methanol after air drying on fixed slides for 30 min.

 

6. 5% normal rabbit serum was blocked.

 

7. Denaturing the nucleic acid at 100 ° C for 5 min.

 

8. After cooling in an ice bath, wash with PBS, add 1 antibody to anti-mouse BrdU monoclonal antibody (working concentration 1:50), and add PBS or serum to the negative control.

 

9. According to the ABC method, hematoxylin or eosin was used for the staining, and the total number of cells and the number of BrdU positive cells in 10 high magnification fields were randomly counted under a microscope to calculate the labeling index (LI).

 

The cells were seeded at a cell number of 1.5 × 105 / ml in a 35 ml diameter dish (a cover glass was placed inside), cultured for 1 day, and synchronized with 0.4% FCS medium for 3 days, so that most cells were in the G0 phase.
